Summary:Circular airlift fermenter system (FAUK) was found to be the best system in producing thermotolerant lipase enzyme from Geobacillus thermodenitrificans (LGT). The results revealed that optimum parameters were, temperature of 65°C, pH of 6.8 and inoculum size at 7.0% (v/v); which were used in the kinetic studies of FAUK system using production medium containing 0.75% (w/v) glucose, 1.25% (w/v) yeast extract, 0.45% (w/v) NaCl and 0.10% (v/v) olive oil (Nik Raikhan, 2003). Glucose at 1.25% (w/v) and air flow rate of 1.00 L/min were the best parameters. Kinetic approach was focused on the effect of glucose concentrations and air flow rates to the cell growth and the production of LGT. The results revealed that specific growth, μ was highest with 1.25% (w/v) and flow rate of 1.0 L/min which was 0.12 hour-1. This cultivation has the lowest doubling time, td that was 5.77 hours, the rate of enzyme formation, dp/dt of 1.73 Uml-1h- 1, a very high rate of specific enzyme production which was recorded as 11700.0 U/g of mass, the rate of glucose used, ds/dt of 0.22 gL-1h-1 and quite a low formation of enzyme per gram, that was 4110.0 U/g of glucose.
