Summary: | Amylases are starch-degrading enzymes that catalyzes the breakdown of glycosidic linkages in low molecular weight molecules like glucose and maltose. To date many studies regarding bacteria have been isolated from the environment that can be used in many industries. The purpose of current research was to screen amylase enzymes activity of bacteria producing amylase from soil samples near Wind cave, Bau and. The submerged fermentation approach is applied to produce amylase at room temperature for 48 hours. It was determined that the amylase activity could be estimated using the supernatant that was obtained. The DNSA approach was utilized to perform an amylase activity assay by determining the amount of reducing sugar. A total of 9 isolates were obtained after performing the spread plate technique. Two of the isolates, FB2 and FB3 displayed a positive response for amylase enzyme by the development of a clear zone of starch hydrolysis on starch agar. It was perceived that isolate FB2 and FB3 exhibiting amylase activity of 6.3024 and 4.2145 U/ml respectively. However, additional exploration is required in order to evaluate the characteristics of the enzyme, as well as its pH stability and the influence of various parameters. |
