Development of Biomarker or DNA Fingerprinting of Pepper Berries from Different Geographical Localities

Genetic variation of Piper nigrum L. depends on revelation of unique strains. Studies reported that amplified fragment length polymorphism (AFLP) technique have been used to detect the genetic diversity and examining genetic relationships in pepper berries (Piper nigrum L. ). Thus, the aim of thi...

Studies reported that amplified fragment length polymorphism (AFLP) technique have been used to detect the genetic diversity and examining genetic relationships in pepper berries (Piper nigrum L. ). Thus, the aim of this research study is to develop biomarkers by extracting the fresh pepper berries DNA and to characterize the unique DNA between locations by looking at their banding patterns; Four geographical localities in Sarawak were chosen which are Serian, Padawan, Bako and Lundu. The pepper berries were collected, and a total of thirteen samples from two locations were successfully extracted from the pepper berries. The DNA from S 1 and Pl were chosen for selective polymerase chain reaction (PCR) with six AFLP primer combinations to identify the most suitable pairs of restriction endonucleases and selective primers. By using 3% agarose gel electrophoresis (AGE), result shows that five primer combinations produced unique bands whereas another one primer combination shows similar and shared fragments between Serian and Padawan samples.
