Q Science (General) This study focused on the isolation and identification of the thermostable bacteria that produce the most highly enzyme activity obtained from Anah Rais Hot Spring. Four selective media have been used namely xylan, starch, CarboxymethyIcellulase (CMC) and skim milk media to isolate specific bacteria from the water sample. The growth of the culture been observed and comparison was done among those different media to select the one that produce the most highly enzyme activity. The enzyme activity was indicated by enzyme degradation of the each media that produced clear haloes around the supernatant (containing the enzyme) of each microorganism. Among those four, the growth of the skim milk media was selected to been studied further onto it morphological and molecular level since it produced the biggest size of halo. Throughout the study, 5 colonies of the proteolytic organisms have been isolated and identified as Sm i Nisseria sp., Sm2 Bacillus sp., Sm3 Lnterohacter sp., Sm4 Klehsiella sp. and Sm5 Streptococcus sp. Biochemical test was carried out in order to identify and differentiate the species. Genomic DNA from each bacteria colony was extracted. However, no further work was carried out due to time limitation.
