An accelerated solvent extraction (ASE) method was developed to extract interest compounds from crude extract of Orthosiphon Stamineus(Benth).ln this study, the interest compound selected is Sinensetin, which is used as anti-cancer agent. The layout of experiment was developed via Statistica 8.0using the Box-Behnken experimental design model. The oil yield and quantification of interest compounds were calculated and the response surface methodology (RSM) was done to compare the result from the experiment and from the statistic. Through the RSM the optimum condition for the extraction process from the Orthosiphon Stamineus crude extract was determined. It was revealed that the value from the model equation was in good agreement with the experimental values with the regression coefficient, RA2, 0.9863.F test also indicated that the model chosen is suitable. 100°C, lOmin and 2extraction cycles were found to be the best and optimum conditions. In other words, highest oil yield, 22.75%, was found in mentioned optimum conditions. HPLC was used to determine concentration of bioactive compound (Sinensetin); however, concentration of SEN was almost negligible in this study.
