In this study, the starch content of the fresh Cladophora sp. collected from natural habitat has been analysed throughout the cultivation period. This study focused on Cladophora sp. cultured in different type of media and culture condition to investigate which is the best for producing the highest biomass, starch and protein in shorter time. The cultivation of the alga with initial 3% w/v (90 g) was conducted in 2 different media: Filtered Tap Water (FTW) and Filtered Sago Effluent (FSE) for 40 days using 3 L working volume. The culturing condition of Cladophora sp. was altered using different types of container: plastic tray and plastic tubing. Approximately 2 g of Cladophora sp. sample was collected every 10 days for analyse. From starch analysis the highest concentration of starch was recorded on day 30 at 0.112 mg/mL.in plastic tubing for tap water and day 40 at 0.159 mg/mL in plastic tray for sago effluent. The highest protein concentration was recorded on day 20 in both Filtered Tap Water and Filtered Sago Effluent using plastic tubing which is 0.032 mg/mL and 0.041 mg/mL respectively.
