Summary:An atlempt on Ibe detection of Marek's disease virus (MDV) in village chickens from selected area in Kuching Division, Sarawak was conducted utilising Polymerase Chain Reaction (PCR) as a technique of detection. Whole blood samples from village chickens were subjected to processes such as deoxyribonucleic acid (DNA) extraction, semi-qualitative and quantitative analysis, PCR amplification and finally agarose gel electrophoresis. The DNA extraction was successful with DNA bands formation on the I % (w/v) agarose gel viewed under UV transilluminator. The quality of the DNA extracts was considered as pure with ratio of A,WA18O in the range of 1.8 to 2.0 fur most samples. The concentration of extracted total genomic DNA from all samples ranged from 38 to 586 ngflr'. However, the PCR amplification process was not successful with unspecific DNA bands formation on the agarose gel and no expected band with the size of 196 bp was observed for the confinnation of the presence ofMD¥.
