The major problem faced in the study is bacterial contamination. It might due to contaminated explant used with bacteria and other microorganisms. Different sterilization methods were tested to overcome this problem. Based on this parameter, the best result was obtained with the combination of 80% (v/v) of ethanol for 20 seconds and 4% (v/v) NaOCI for 3 minutes (Method 2). Further study were carried out to investigate the effect of cytokinin on shoot initiation in three different concentration of BAP/Kn/2ip and constant NAA/IAA/2 BAP/Kn/2ip,4-D. Result shown that MS media supplemented with 0.5mg/L IAA + 1.0mg / 1 2ip give shoot after 2 weeks of culture.
