Summary:Pollen viability is important for the success of sexual reproduction in plants. Based on viability, pollen quality is assessed in order to improve fruit quality and productivity. The study focus at evaluating the pollen viability of three different guava cultivars in Malaysia. The colorimetric method of estimation of pollen viability was done by using 2,3,5-triphenyl tetrazolium chloride (TTC) and Iodine Potassium Iodide (IKI) solution. The observation for the viability of pollen were made at x40 magnification using a light microscope. IKI staining has higher pollen viability percentage (97.93 - 99.59%) in all guava species compared to TTC (2.29% - 59.26%). The results of the Mann-Whitney test showed that the differences in pollen viability under IKI and TTC staining were significant at p < 0.05, z = -6.685. The highest viable pollen were recorded from Lohan, followed by Pink guava heirloom and the least from Kim Choo red. However, Kruskal-Wallis analysis at p < 0.05 revealed that the distribution of pollen viability among cultivars was not significantly different. According to these observations, viability obtained by staining methods are easier and faster, but may frequently yields erroneous positive findings. Thus, other test such as in vitro germination method may be useful to study the occurrence of pollen viability.
